Question
I'm a duty holder at my office and have just noticed next weeks day off which should be Saturday is now Thursday surely duty holders days off can't be changed just too meet operational needs......can they?

No they can't, I wouldn't say anything, when you turn up on Thursday and they ask why just say I'm a duty holder so my day off is rolling, why would I need to look at the duty rota?
Then walk off and carry on as normal, but that's just me!

Or take the Thursday and the Saturday off.

In an office with reserves it's not very likely to happen all that often but if it did i would seek advice before i refused point blank.
It would depend on the circumstances,how it was put to you and the length of notice given as to whether it could be considered a reasonable request.

TBH I'd rather have a Thursday off than a Saturday

Stand your ground FFS 
unless you dont mind having your day changed then tell your Dom that its not on you have a rotating R/Day as agreed in a revision,stick a reserve on the Thursday or ask some one whos day off it is that day if they want to come in and work it
